GETTING PREPARED IN THE LABORATORY FOR LC-MS WORK
-
Filters we use for sample prep are Millex-HV from
Millipore 4mm, Cat no SLHV R04 NL. If you
have enough sample volume: 13 mm ones, same quality
-
Volumetric Pyrex
flasks: volumes 10 ; 20; 100 ml minimum (typically 5 each).
-
Measuring glass cylinders 50; 100; 250; 500;
1000 minimum (typically 2 each)
-
Most Important: Boro Silicate Pyrex
good grade solvents' bottles: 250, 500. 1000
-
Vortex,
-
Ultrasonic bath to clean filters and source's cone (typically
once a month or more)
-
Mandatory : Pyrex beakers sizes 600 ml
for source cleaning, than 200; and 100.
-
Proper Gloves (clean)
-
Kimwipes
- Reserve of Syringes GAS Tight Hamilton: sizes 250ul,
5000ul and/or 10000ul for the infusion pump
-
Peek tubing and nuts
-
For sample prep 1; 2.5; 5 ml plastic disposable
syringes (or disposable pipettes) are OK, as they are not
contain the samples for a long time.
-
Accurate thermometer to measure room temperature - sometimes
needed for the extra mass accuracy verification.
Important note: DO NOT USE PLASTICS CONTAINERS AT ANY
POINT OF SAMPLE OR MOBILE PHASE PREPARATION (preservatives).
Pyrex is the best, as it does not leach out any ions or
preservatives/
Buffers and solvents grades:
1. Extra pure acetonitril, methanol and water LC-MS
and/or UPLC grade (test them before for impurities)
2. Extra pure formic acid (check that the brand is used
elsewhere in LC-MS)
3. Extra pure ammonium formate and ammonium acetate
(volatile buffers for LC-MS)
3. Extra pure ammonia for LC-MS.
